  The GO Test (The Guanosine Oxidation Test)



  dG ⇒ 8-oxo-dG oxidation index indicates the probability of the oxidative induction of dG intra- and/or extracellularly. Therefore, the GO test can predict the influence of chemical substances in test samples exposed to the gene. The GO test based on a patent (International Publication No. WO 2001069235 A1) was developed at the Kyushu University and modified to accommodate various needs.
  1. Correlation with the conventional test
  2.   The correspondence of the result of the GO test with that of the Ames test, a conventional mutagenicity test, is >90% for standard mutagenic chemicals. In addition, 72% of chemicals classified as carcinogens showed significant oxidative reactions in the GO test. The lower correspondence of carcinogenicity than of mutagenicity may be explained by a mechanism other than mutation. In all cases, the GO test is very useful for screening before the conventional test because it can simultaneously access many samples.

  3. Fulfillment of 3 Rfs (Replacement, Reduction, and Refinement)
  4.   One of the procedures in the GO test requires neither living cells nor experimental animals. We can estimate biological toxicity resulting from the physicochemical influence on nucleic acids, particularly dG, in an aqueous solution. Therefore, it is cruelty-free test.

  5. Applicable domain
  6.   It is applicable to materials containing a variety of molecules such as food, natural crude drugs, cosmetics, and environmental media (water, air, and soil) besides simple chemicals that can be dissolved in water. There is a sharp difference from the conventional mutagen test, the Ames test. The result does not depend on the nutritional components in samples, particularly essential amino acids, because it is not necessary to keep a microbe alive. Moreover, foods can be evaluated as final products and not raw materials, which processing and addition, sterilization, bag filling ended in is a big advantage.

  7. Evaluation of antioxidative effects using the GO index
  8.   The GO index reflects the generation of hydroxyl radicals, which are very strong reactive oxygen species, in an aqueous solution containing the test sample in vitro. In addition, we can also evaluate the antioxidative potential of the target materials by the suppression of the generation of hydroxyl radicals, indicated by the inhibition of the GO index when potassium bromate, a representative hydroxyl radical inducer, is added.
